Cilji:

Študent se bo seznanil z načinom, kako postavljati vprašanja o strukturi, dinamiki in zvijanju (folding) proteinov in to iz eksperimentalnega stališča. Bo tudi zmožen kritično presoditi, kako so te tri komponente medsebojno povezane.

Vsebina:

Označevanje proteinov (Labelling): kako napravimo protein občutljiv za NMR metode

Strategije asigniranja resonanc: katero jedro (proton) določa posamezni signal

Določanje strukture v raztopini: metodologija kako dobiti makromolekulske structure iz NMR podatkov

Rezidualne dipolne sklopitve: ocena strukture in izboljšanje natančnosti

Interakcije med proteini: NMR metode za študij interakcje med proteini

Vezava ligandov: NMR metode za študij vezave ligandov

Izmenjava vodikov (z devterijem): proučevanje stanj, ki obstajajo v nizki populaciji

Ne-zvita stanja proteinov: NMR in izmenjava med konformacijskimi stanji

Študij dinamike proteinov: gibanja znotraj zvitih ali odvitih makromolekul
